采用RT-PCR方法自犬瘟热病毒Lederle株基因组RNA中扩增出H基因(1 911 bp)和N基因(1 707 bp)片段,序列测定和分析表明与Onderstepoort株、CDV3株等疫苗毒株的同源性在95%以上,而与野外分离株的同源性介于90%~95%;以获得的H,N全基因片段为模板,分别扩增H基因近3′端996 bp的基因片段以及N基因中间高度保守区562 bp的基因片段,经克隆测序后分别插入原核表达载体pET-32a中进行表达,结果显示H片段和N片段分别表达大小约为57 kD和37 kD的融合蛋白,Western blotting检测结果显示,表达蛋白均可与CDV抗血清发生阳性反应,提示原核表达产物具有与CDV抗血清结合的免疫反应性。 相似文献

根据鸡传染性贫血病毒(CIAV)基因组序列,设计一对引物扩增基因组上nt358~nt1042之间684bp DNA片段。在CIAV感染MDCC-MSBl细胞系中提取核酸为模板可扩增出相应DNA片段,且以感染细胞核酸为模板反应敏感性可达1fg,而以其他病原核酸为模板扩增结果均为阴性。应用该PCR方法对实验感染鸡肝脏、脾脏、胸腺、肾脏、法氏囊、骨髓、白细胞、泄殖腔棉拭子等不同样品进行检测,均可扩增出相应DNA片段。证明我们建立的PCR方法敏感性特异性强,可用于临床感染不同组织CIAV的检测。 相似文献

Summary Plant viruses cause considerable losses to crops and none of the three current approaches to disease control are likely to give the long-term answer. The new concept of non-conventional protection, in which the expression of a viral or virus-related sequence in the plant genome interferes with the virus infection cycle, holds considerable promise for designing new resistance or protection genes. A series of targets in the viral genome is identified and a range of mechanisms for attacking those targets is discussed. There are several current systems which have been proved to give protection, at least to a certain extent, and many systems which are being researched upon for the future. Some of these are described to give a picture of the current situation and of the thinking for the future. The problems of field deployment of the transgenic plants are discussed, especially those associated with the risk to the environment. Various questions which molecular biologists and plant breeders will have to consider include what are desirable characters to have in protection genes and how these new genes should be deployed. 相似文献

Summary Resistance to lettuce mosaic virus (LMV) is derived either from cv. Gallega (g gene) or the wild accession PI251245 (mo gene). Previous studies indicated that these two genes were identical. Breeders in Europe produced numerous resistant cultivars utilisingg while in the USAmo was used. The resistance has been effective for over 20 years. However, recently there have been reports of LMV isolates causing unusually severe and sometimes necrotic symptoms on cultivars with these resistance genes. Investigations of these severe isolates have distinguished three new pathotypes in addition to the common pathotype (II) and identified a novel dominant gene for resistance. Themo/g genes confer resistance to pathotypes I and II but pathotype III possesses virulence for cultivars withg but not for those withmo. These two genes are therefore not identical but are probably either closely linked genes or alleles. Pathotype IV possesses virulence for all lettuce lines so far tested. Some isolates of this pathotype are seed transmitted in cultivars possessingmo org and have caused severe crop losses in southern France. Summary Gene pyramiding in Phaseolus vulgaris is being utilized to develop more effective resistance to the temperature-insensitive-necrosis-inducing (TINI) strains of Bean Common Mosaic Virus (BCMV) present in the USA. Our data indicate that contrary to previous work, the bc-3 gene is effective against these strains in the absence of the strain unspecific bc-u gene in genotypes possessing the I gene. The epistatic bc-3 gene interferes with traditional efforts to pyramid the other recessive resistance genes by masking their activity. Indirect selection based on markers linked to the other recessive resistance genes would likewise be ineffective without the ability to also select for the bc-u gene which is required for expression of the bc-2
2 gene in germplasm carrying the I gene. Because the most resistant genotype (I, bc-u, bc-I

Capsid size distributions of viral communities in the floodwater of a Japanese paddy field under a long-term fertilizer trial were surveyed during the rice cultivation period by using transmission electron microscopy. The capsid size distributions were monophasic, and the median values fell within the range of 50-70 nm. The quartile intervals were narrow from either 40-50 nm or 50-60 nm to either 60-70 nm or 70-80 nm for most samples. There was no clear seasonal variation in the capsid size distribution of viral communities. The difference in size distributions among different fertilizer plots was also not found. Background: Historically, histiocytic ulcerative (HUC) (or granulomatous) colitis of Boxer dogs was considered an idiopathic immune-mediated disease with a poor prognosis. Recent reports of dramatic responses to enrofloxacin and the discovery of invasive Escherichia coli within the colonic mucosa of affected Boxer dogs support an infectious etiology.
Hypothesis: Invasive E. coli is associated with colonic inflammation in Boxer dogs with HUC, and eradication of intramucosal E. coli correlates with clinical and histologic remission.
Animals: Seven Boxer dogs with HUC.
Methods: Prospective case series. Colonic biopsies were obtained at initial evaluation in 7 dogs, and in 5 dogs after treatment with enrofloxacin. Biopsies were evaluated by standardized histopathology, and fluorescence in situ hybridization (FISH) with probes to eubacteria and E. coli .
Results: Intramucosal E. coli was present in colonic biopsies of 7/7 Boxers with HUC. Clinical response was noted in all dogs within 2 weeks of enrofloxacin (7±3.06 mg/kg q24 h, for 9.5±3.98 weeks) and was sustained in 6 dogs (median disease-free interval to date of 47 months, range 17–62). FISH was negative for E. coli in 4/5 dogs after enrofloxacin. E. coli resistant to enrofloxacin were present in the FISH-positive dog that relapsed.
Conclusions and Clinical Relevance: The correlation between clinical remission and the eradication of mucosally invasive E. coli during treatment with enrofloxacin supports the causal involvement of E. coli in the development of HUC in susceptible Boxer dogs.
